PAIN-NSAID MECHANISMS
SUMMARY
1. COX-1: responsible for homeostatic functions such as platelet aggregation & maintenance of UGIT mucosal integrity by producing protective prostaglandins.
2. COX-2: inducible cytokine that produces inflammation & pain.
3. Conventional (non-specific) NSAIDs: first-line treatment for analgesia and the treatment of inflammatory conditions (OA, RA, various musculoskeletal-related conditions).
4. Oral COX-2 inhibitors approved by FDA: celecoxib, meloxicam spares COX-1 at selected doses.
